High-throughput screening (HTS) is a key tool which has been leveraged for decades in the identification of novel leads for early drug discovery programs. Over these decades the process and associated technology have advanced significantly, enabling highly miniaturized assays for very challenging targets. Jonathan O’Connell has over 27 years of experience in HTS and will cover the background, share some key elements of building a successful HTS capability, and demonstrate how Dewpoint is running up to a half million compounds through purely phenotypic assays in 1536 well plates in less than a month. HTS is a fully-integrated element in Dewpoint’s discovery platform, leveraged not only to discover novel condensate-modifying drugs (c-mods), but also to rapidly advance leads through lead optimization.
